The A635 Mancunian Way has been closed in both directions in the early hours of Tuesday morning.
Photos from the scene show multiple emergency service vehicles are in attendance.
The major route, part of inner ring road, is shut between between London Road and Fairfield Street. Traffic is being diverted on to London Road and motorists are advised to approach the area with caution.
Traffic service Inrix are reporting that the closure is due to a collision.
Transport for Greater Manchester said on Twitter in the early hours of Tuesday morning: “ROAD CLOSURE. Mancunian Way, both directions between London Road and Fairfield Street.
“Traffic is being diverted off the Mancunian Way via London Road. Please approach with caution.”
The Mancunian Way forms part of the Manchester-Salford inner ring road to the south of the city centre.
The closure has been confirmed by Transport for Greater Manchester on Twitter.
